Don't promote your ISA Server to be a DC, ISA will not operate
correctly. AD will not operate correctly. You will leave yourself open
to compromise due to the many holes you will have to poke through your
firewall.

My ISAServer2004 Enterprise is a member server of my domain and the
Configuration Storage Server.
I had "User sets" defined with domain users and domain groups members,
but in the rules "From/Exception" fields, "Users Sets" are not available
Why?
I need to promote the ISA server as an Auxiliar Domain Controler for my
domain (it make sense)?
