I haven't actually done it, but...  From what I read you do it the
opposite, you grant permission to the SMTP Virtual Server and just leave
that user out.  Take a look here:

This is on an Exchange 2003 server. Can I set one particular user to be
able to send and receive e-mail from only other addresses within our
company. I want to restrict the ability to send and receive externally
for just one user.
