Are you thinking of doing NTFS defragging? If so, don't.

If you are doing an offline defrag of an exchange DB, then is there a
good reason to do it (like one of the DBs has a large amount of free
space after an online defrag)?

Anyone know of anyproblems defragging harddrives on an Exchange Server?
I know to perform maintenance on the mailbox store you must dismount
store but I am wanting to defrag entire harddrives on my exchange
server.

Thanks Bruce
