While backing up the System State is nice and useful. I also like to run on
a nightly basis, LDIFDE and export the groups and users to LDF files. I
include these files in the nightly backup that also backs up the Information
Stores. That gives me a good base to recover from if I can't use the System
State for some reason.

For the first question: No, But you should backup also the system state.
For the second question: Ntbackup is backing up the whole information
store you cannot recover a single mailbox. Only third party backup
software does this. I use backup Exec for backing up my Exchange Server.
