Yes. If you have a keyboard that also has a sampler on it, like the motif, triton, or fantom, for example, you can load samples into the keyboard and play them from there. You can also do this through programs like v sampler or SFz in Sonar. You'll have to keymap the instruments though, and that can become a cumbersome process. I haven't keymapped instruments yet myself, so if you're interested in doing that, someone else on the list may be able to help you.

Thanks, Omar! but what I want to know is, could I play them on the midi keyboard like an instrument? For example, I'd like a sample of a guitar, so I buy or download one, import it and whatever else I'd have to do, then play my midi keyboard, and each note would sound like the note of a guitar. Is that how it can work? I just want to clarify.
