Can anyone recommend a freeware/open source document management system for
use in an organisation of about 100 people?

I can't offer a recommendation, but can help you to identify some systems.
If you go to <http://sourceforge.net/>, enter document management in the
search field, and click Search, you will be presented with a list of results
that contains some possibilities. The following may be of interest from just
the first page of results:

Owl Intranet Knowledgebase - a multi user document repository
(knowledgebase), document manager and/or document management system written
in PHP for publishing of files/documents onto the web for a corporation,
small business, group of people, or just for yourself.

OpenDocMan - a web based document management system designed to comply with
ISO 17025 and OIE standard for document management. It features fine grained
control of access to files...

TUTOS - the ultimate team organization software, a webbased groupware or
ERP/CRM system to manage events/calendars, addresses, teams, projects,
tasks, bugs, mailboxes, documents.
