If you are booting from USB-FDD then DOS will work with the floppy as
normal. The BIOS should support booting from USB-FDD, if not you are out
of luck with USB-FDD booting.
Otherwise if you are booting from harddisk/cd then it's not sure
whenever you can access the USB-FDD, seldom the BIOS offers an option
USB legacy support for mass storage, see

I did never keep care if DOS USB drivers also support USB-FDD, you have
to check it out yourself.

If all cords break you can still use either eltorito floppy emulation or
grub4dos (memdisk like) device emulation and images.
