Shannon, I have the same scanner and did a lot of resolution / sharpness
testing. If you want me to send you my tests charts will do but there are 6
or 7 emails to keep download time low.
Conclusion: Better sharpness and resolution using film holder setting but
not a lot. Exposure is increased. Best point of focus even with depth of
field is either film against the glass or inverted within the film base
thickness. So
either emulsion up or emulsion down works. But for my.. scanner don't use
the plastic film holders.
Even with the AN glass I got Newton rings so I hold the MF film down with
lead weights on the edges. I have not tried 4x5 negs.

You can get a fixed height one or a variable height one. I use the epson
4990 scanner, and I wonder if anybody knows whether the fixed height one
is really at the right height for scanning on that scanner, or if you
really need the variable height one. The fixed height one seems simpler.
