What virtualization software do you use?

I'm not sure if I fully understood your intentions. Does the guest file
system reside on a separate Volume on the host? Or have you set up LVM
inside of the guest itself? In other words, is the guest aware of the
host's volumes and does "lvdisplay" from inside the guest give you any
output?

I assume that this is not the case and you can not create snapshot
volumes from inside the guest then, as the volumes are managed by the
host itself. However, if the host can access the file system of the
guest directly (e.g. if it's mounted on the host side), you could try to
execute the backup script from the host, but have to make sure that it
accesses the mysql server running inside the guest (to issue the flush
and lock commands). The existing script should already provide you with
the options to do that (by modifying the path names and connection
options), by either connecting to the TCP port or the socket file
located inside the guest file system.

Let us know if it worked!

Bye,
        LenZ
