What is the tube made of?

I'll assume sonotube.

As a quick thought, a latch that compresses the pieces together will
just be pushing cardboard to cardboard and this couldn't be stiff
enough.

Perhaps some sort of "ears" that are firmly permamently bolted to one
half and then bolted/unbolted to the other half might be stonger.
Look at the split tubes from Discovery to see what I am talking about
(http://www.discovery-telescopes.com/discovery/pdhq.html). Here's
(http://www.cloudynights.com/item.php?item_id=549) and example of a
Meade modified this way.
